NETs are formed when neutrophils externalize their decondensed chromatin together with content from their azurophilic granules. In addition to their role in defense against microbes, NETs have been implicated as mediators of pathology in sterile inflammation, such as cancer and autoimmunity, and their potential as therapeutic targets is actively explored. However, targeting of NETs is challenging since the beneficial effects of their removal need to be balanced against the potential harmful loss of their function in microbial defense. Moreover, depending on the stimuli or species, NETs can be formed via distinct mechanisms and are not always made up of the same components, making direct comparisons between various studies challenging. This review focuses on the role of NETs in cancer-associated pathology, such as thrombosis, organ dysfunction, and metastasis. Different strategies to target NETs, by either preventing their formation or degrading existing ones, are also discussed.", "doi": "10.1152/physrev.00062.2021", "pmid": "35951483", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC9576172"}], "notes": [], "created": "2026-08-21T12:28:08.476Z", "modified": "2026-08-21T12:28:08.665Z"}, {"entity": "publication", "iuid": "102f3b1aad7d4b81a1d0666f2986612f", "links": {"self": {"href": "https://publications-affiliated.scilifelab.se/publication/102f3b1aad7d4b81a1d0666f2986612f.json"}, "display": {"href": "https://publications-affiliated.scilifelab.se/publication/102f3b1aad7d4b81a1d0666f2986612f"}}, "title": "Neutrophil extracellular traps promote cancer-associated inflammation and myocardial stress.", "authors": [{"family": "Cedervall", "given": "J", "initials": "J", "orcid": "0000-0002-9251-3318", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/d0c756e54d634faea619cee35ea70500.json"}}, {"family": "Herre", "given": "M", "initials": "M"}, {"family": "Dragomir", "given": "A", "initials": "A", "orcid": "0000-0003-2777-8114", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/0097cdbab8cf450688184aef60a6f8b3.json"}}, {"family": "Rabelo-Melo", "given": "F", "initials": "F"}, {"family": "Svensson", "given": "A", "initials": "A"}, {"family": "Th\u00e5lin", "given": "C", "initials": "C"}, {"family": "Rosell", "given": "A", "initials": "A"}, {"family": "Hjalmar", "given": "V", "initials": "V"}, {"family": "Wall\u00e9n", "given": "H", "initials": "H"}, {"family": "Lindman", "given": "H", "initials": "H"}, {"family": "Pejler", "given": "G", "initials": "G"}, {"family": "Hagstr\u00f6m", "given": "E", "initials": "E", "orcid": "0000-0003-3221-0144", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/0b98b623b2ff42fdbdff3175f37b322e.json"}}, {"family": "Hultstr\u00f6m", "given": "M", "initials": "M"}, {"family": "Larsson", "given": "A", "initials": "A"}, {"family": "Olsson", "given": "A K", "initials": "AK"}], "type": "journal article", "published": "2022-03-14", "journal": {"title": "Oncoimmunology", "issn": "2162-402X", "volume": "11", "issue": "1", "pages": "2049487", "issn-l": "2162-4011"}, "abstract": "Cancer is associated with systemic pathologies that contribute to mortality, such as thrombosis and distant organ failure. The aim of this study was to investigate the potential role of neutrophil extracellular traps (NETs) in myocardial inflammation and tissue damage in treatment-na\u00efve individuals with cancer. Mice with mammary carcinoma (MMTV-PyMT) had increased plasma levels of NETs measured as H3Cit-DNA complexes, paralleled with elevated coagulation, compared to healthy littermates. MMTV-PyMT mice displayed upregulation of pro-inflammatory markers in the heart, myocardial hypertrophy and elevated cardiac disease biomarkers in the blood, but not echocardiographic heart failure. Moreover, increased endothelial proliferation was observed in hearts from tumor-bearing mice. Removal of NETs by DNase I treatment suppressed the myocardial inflammation, expression of cardiac disease biomarkers and endothelial proliferation. Compared to a healthy control group, treatment-na\u00efve cancer patients with different malignant disorders had increased NET formation, which correlated to plasma levels of the inflammatory marker CRP and the cardiac disease biomarkers NT-proBNP and sTNFR1, in agreement with the mouse data. Altogether, our data indicate that NETs contribute to inflammation and myocardial stress during malignancy. These findings suggest NETs as potential therapeutic targets to prevent cardiac inflammation and dysfunction in cancer patients.", "doi": "10.1080/2162402X.2022.2049487", "pmid": "35309730", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC8928831"}, {"db": "pii", "key": "2049487"}], "notes": [], "created": "2026-08-21T12:05:32.854Z", "modified": "2026-08-21T12:05:32.984Z"}]}
