Here, we show that the ErbB3 (HER3) receptor sorts internalized Integrin \u03b21 and the transferrin receptor, for endocytic recycling, in a manner that does not require ligand-induced ErbB3 signaling in breast epithelial cells. Loss of ErbB3 abrogates recycling of Integrin \u03b21, likely from a Rab4-positive compartment, and redirects it toward lysosomal degradation or secretion as an extracellular vesicle (EV) cargo. ErbB3 depletion impairs the collective migration of breast epithelial cell sheets, coinciding with reduced cell-surface levels of Integrin \u03b21 and increased release of Integrin \u03b21-containing EVs. In contrast, EVs secreted from ErbB3-depleted cells enhance the motility of wild-type cells. Mechanistically, ErbB3 promotes assembly of the Arf6-GGA3-Rabaptin5 endosomal sorting complex to facilitate early recycling and suppress EV release. Organized by eight National EV Societies under the MOVE initiative, the event gathered over 280 attendees from 28 countries, promoting collaboration and scientific exchange. The symposium featured eight keynote lectures, 48 oral and 126 poster presentations, and sessions dedicated to EV-related tools and industry innovations. The scientific program was structured around seven core themes: EV biogenesis and signal transmission, roles of EVs in health and disease, EV-based biomarkers, interspecies communication, novel EV preparation and analysis techniques, therapeutic and regenerative applications, and the manufacturing of native and engineered EV products. Supported by 18 sponsors and the Ministry of science, technological development and innovation of the Republic of Serbia, the symposium also highlighted the MOVE Fellowship Program and offered rich networking opportunities. Although therapeutic approaches against TC have improved in recent years, the survival rate remains low, and severe adverse effects are commonly reported. However, unexplored alternatives based on natural compounds, such as lysicamine, an alkaloid found in plants with established cytotoxicity against breast and liver cancers, offer promise. Therefore, this study aimed to explore the antineoplastic effects of lysicamine in papillary TC (BCPAP) and ATC (HTH83 and KTC-2) cells. Lysicamine treatment reduced cell viability, motility, colony formation, and AKT activation while increasing the percentage of necrotic cells. The absence of caspase activity confirmed apoptosis-independent cell death. Necrostatin-1 (NEC-1)-mediated necrosome inhibition reduced lysicamine-induced necrosis in KTC-2, suggesting necroptosis induction via a reactive oxygen species (ROS)-independent mechanism. Additionally, in silico analysis predicted lysicamine target proteins, particularly those related to MAPK and TGF-\u03b2 signaling.
