Here we show that specific snoRNAs are required for the activation of immune response genes and for survival during viral infections in Drosophila melanogaster. We have studied snoRNA:U3:9B, a chromatin-associated snoRNA that binds to a large number of protein coding genes, including immune response genes. We have used CRISPR/Cas9 to delete snoRNA:U3:9B and study its function in vivo. SnoRNA:U3:9B-deficient larvae are viable but failed to develop into pupae when challenged by expression of a Sindbis virus replicon. SnoRNA:U3:9B is localized to immune genes in vivo and the chromatin decompaction and gene activation typically observed at immune genes following infection are abolished in snoRNA:U3:9B-deficient larvae, which suggests that this snoRNA acts locally to regulate chromatin accessibility. Mechanistically, snoRNA:U3:9B is required for the recruitment of the chromatin remodeler Brahma to a set of target immune genes. In summary, these results uncover an antiviral defense mechanism that relies on a snoRNA for the recruitment of a chromatin remodeling factor to immune genes to facilitate immune gene activation.", "doi": "10.1093/nar/gkaf715", "pmid": "40737091", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC12309367"}, {"db": "pii", "key": "8219220"}], "notes": [], "created": "2026-08-21T12:11:18.732Z", "modified": "2026-08-21T12:11:18.810Z"}, {"entity": "publication", "iuid": "7faaa53e449942c0958fdb8ef6555a50", "links": {"self": {"href": "https://publications-affiliated.scilifelab.se/publication/7faaa53e449942c0958fdb8ef6555a50.json"}, "display": {"href": "https://publications-affiliated.scilifelab.se/publication/7faaa53e449942c0958fdb8ef6555a50"}}, "title": "Separation of transcriptional repressor and activator functions in Drosophila HDAC3.", "authors": [{"family": "Tang", "given": "Min", "initials": "M", "orcid": "0000-0003-3492-0262", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/e3082950b0a4495c8ce7575fc7e6f065.json"}}, {"family": "Regadas", "given": "Isabel", "initials": "I"}, {"family": "Belikov", "given": "Sergey", "initials": "S"}, {"family": "Shilkova", "given": "Olga", "initials": "O"}, {"family": "Xu", "given": "Lei", "initials": "L"}, {"family": "Wernersson", "given": "Erik", "initials": "E"}, {"family": "Liu", "given": "Xuewen", "initials": "X", "orcid": "0000-0001-5458-0731", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/77d2b082c52f49fb9803ca1a789987c4.json"}}, {"family": "Wu", "given": "Hongmei", "initials": "H", "orcid": "0000-0003-2232-6872", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/0c30767339ba4a738b177a70285fac58.json"}}, {"family": "Bienko", "given": "Magda", "initials": "M"}, {"family": "Mannervik", "given": "Mattias", "initials": "M", "orcid": "0000-0003-4999-9655", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/4f97a73be65f402b8d27db787616945d.json"}}], "type": "journal article", "published": "2023-08-01", "journal": {"title": "Development", "issn": "1477-9129", "volume": "150", "issue": "15", "issn-l": "0950-1991"}, "abstract": "The histone deacetylase HDAC3 is associated with the NCoR/SMRT co-repressor complex, and its canonical function is in transcriptional repression, but it can also activate transcription. Here, we show that the repressor and activator functions of HDAC3 can be genetically separated in Drosophila. A lysine substitution in the N terminus (K26A) disrupts its catalytic activity and activator function, whereas a combination of substitutions (HEBI) abrogating the interaction with SMRTER enhances repressor activity beyond wild type in the early embryo. We conclude that the crucial functions of HDAC3 in embryo development involve catalytic-dependent gene activation and non-enzymatic repression by several mechanisms, including tethering of loci to the nuclear periphery.", "doi": "10.1242/dev.201548", "pmid": "37455638", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC10445730"}, {"db": "pii", "key": "323882"}], "notes": [], "created": "2026-08-20T12:38:23.231Z", "modified": "2026-08-20T12:38:23.426Z"}, {"entity": "publication", "iuid": "39835aacb6a54f02acecd0a2d532c301", "links": {"self": {"href": "https://publications-affiliated.scilifelab.se/publication/39835aacb6a54f02acecd0a2d532c301.json"}, "display": {"href": "https://publications-affiliated.scilifelab.se/publication/39835aacb6a54f02acecd0a2d532c301"}}, "title": "Separation of transcriptional repressor and activator functions in HDAC3", "authors": [{"family": "Tang", "given": "Min", "initials": "M"}, {"family": "Regadas", "given": "Isabel", "initials": "I"}, {"family": "Belikov", "given": "Sergey", "initials": "S"}, {"family": "Shilkova", "given": "Olga", "initials": "O"}, {"family": "Xu", "given": "Lei", "initials": "L"}, {"family": "Wernersson", "given": "Erik", "initials": "E"}, {"family": "Liu", "given": "Xuewen", "initials": "X"}, {"family": "Wu", "given": "Hongmei", "initials": "H"}, {"family": "Bienko", "given": "Magda", "initials": "M", "orcid": "0000-0002-6499-9082", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/4b8da7a1f8524d0e812c83b1f5e20ef6.json"}}, {"family": "Mannervik", "given": "Mattias", "initials": "M", "orcid": "0000-0003-4999-9655", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/4f97a73be65f402b8d27db787616945d.json"}}], "type": "posted-content", "published": "2022-06-11", "journal": {"issn-l": null}, "abstract": null, "doi": "10.1101/2022.06.11.495646", "pmid": null, "labels": [], "xrefs": [], "notes": [], "created": "2026-08-20T10:11:46.916Z", "modified": "2026-08-20T10:11:46.967Z"}]}
