This work studies these properties by varying the structure of the constituting lipids in order to influence their interaction with high-density lipoprotein (HDL) particles. Various fluorescence-based techniques were applied to study lipid domains, membrane order, and the overall lateral as well as the molecule-internal glycerol region mobility in HDL-membrane interactions (i.e., binding and/or cargo transfer). The analysis of interactions with HDL particles and various lipid phases revealed that both fully fluid and some gel-phase lipids preferentially interact with HDL particles, although differences were observed in protein binding and cargo exchange. Both interactions were reduced with ordered lipid mixtures containing cholesterol. To investigate the mechanism, membranes were prepared from single-lipid components, enabling step-by-step modification of the lipid building blocks. On a biophysical level, the different mixtures displayed varying stiffness, fluidity, and hydrogen bond network changes. Increased glycerol mobility and a strengthened hydrogen bond network enhanced anchoring interactions, while fluid membranes with a reduced water network facilitated cargo transfer. In summary, the data indicate that different lipid classes are involved depending on the type of interaction, whether anchoring or cargo transfer.", "doi": "10.3390/membranes14120261", "pmid": "39728711", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC11677176"}, {"db": "pii", "key": "membranes14120261"}], "notes": [], "created": "2026-08-20T13:42:17.405Z", "modified": "2026-08-20T13:42:17.678Z"}, {"entity": "publication", "iuid": "e51accc010244a71aa40f5260f377308", "links": {"self": {"href": "https://publications-affiliated.scilifelab.se/publication/e51accc010244a71aa40f5260f377308.json"}, "display": {"href": "https://publications-affiliated.scilifelab.se/publication/e51accc010244a71aa40f5260f377308"}}, "title": "Lipoprotein Particles as Shuttles for Hydrophilic Cargo.", "authors": [{"family": "Weber", "given": "Florian", "initials": "F", "orcid": "0000-0002-8946-1377", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/bb14aeb3d7d742d3a657cf49c0e48900.json"}}, {"family": "Axmann", "given": "Markus", "initials": "M", "orcid": "0000-0001-9818-9734", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/12e33cef083245ae9d9e007da19c174e.json"}}, {"family": "Horner", "given": "Andreas", "initials": "A", "orcid": "0000-0002-5328-1745", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/a49f7611927b45b48dc88329dc762bb7.json"}}, {"family": "Schwarzinger", "given": "Bettina", "initials": "B"}, {"family": "Weghuber", "given": "Julian", "initials": "J", "orcid": "0000-0001-6312-4666", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/4922df367ceb470997fdab80877d3d3c.json"}}, {"family": "Plochberger", "given": "Birgit", "initials": "B", "orcid": "0000-0003-2733-9947", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/b87991abbc944513910f6d9fe229af70.json"}}], "type": "journal article", "published": "2023-04-28", "journal": {"title": "Membranes (Basel)", "issn": "2077-0375", "volume": "13", "issue": "5", "issn-l": "2077-0375"}, "abstract": "Lipoprotein particles (LPs) are excellent transporters and have been intensively studied in cardiovascular diseases, especially regarding parameters such as their class distribution and accumulation, site-specific delivery, cellular internalization, and escape from endo/lysosomal compartments. The aim of the present work is the hydrophilic cargo loading of LPs. As an exemplary proof-of-principle showcase, the glucose metabolism-regulating hormone, insulin, was successfully incorporated into high-density lipoprotein (HDL) particles. The incorporation was studied and verified to be successful using Atomic Force Microscopy (AFM) and Fluorescence Microscopy (FM). Single-molecule-sensitive FM together with confocal imaging visualized the membrane interaction of single, insulin-loaded HDL particles and the subsequent cellular translocation of glucose transporter type 4 (Glut4).", "doi": "10.3390/membranes13050471", "pmid": "37233532", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC10222575"}, {"db": "pii", "key": "membranes13050471"}], "notes": [], "created": "2026-08-21T13:04:13.619Z", "modified": "2026-08-21T13:04:13.768Z"}]}
