In this study, we performed genome sequencing for 123 sighthounds, including one breed from Africa, six breeds from Europe, two breeds from Russia, and four breeds and 12 village dogs from the Middle East. We gathered public genome data of five sighthounds and 98 other dogs as well as 31 gray wolves to pinpoint the origin and genes influencing the morphology of the sighthound genome. Population genomic analysis suggested that sighthounds originated from native dogs independently and were comprehensively admixed among breeds, supporting the multiple origins hypothesis of sighthounds. An additional 67 published ancient wolf genomes were added for gene flow detection. Results showed dramatic admixture of ancient wolves in African sighthounds, even more than with modern wolves. Whole-genome scan analysis identified 17 positively selected genes (PSGs) in the African population, 27 PSGs in the European population, and 54 PSGs in the Middle Eastern population. None of the PSGs overlapped in the three populations. Pooled PSGs of the three populations were significantly enriched in \"regulation of release of sequestered calcium ion into cytosol\" (gene ontology: 0051279), which is related to blood circulation and heart contraction. In addition, ESR1, JAK2, ADRB1, PRKCE, and CAMK2D were under positive selection in all three selected groups. This suggests that different PSGs in the same pathway contributed to the similar phenotype of sighthounds. We identified an ESR1 mutation (chr1: g.42,177,149 T > C) in the transcription factor (TF) binding site of Stat5a and a JAK2 mutation (chr1: g.93,277,007 T > A) in the TF binding site of Sox5. Functional experiments confirmed that the ESR1 and JAK2 mutation reduced their expression. Our results provide new insights into the domestication history and genomic basis of sighthounds.", "doi": "10.1093/molbev/msad158", "pmid": "37433053", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC10401622"}, {"db": "pii", "key": "7222862"}], "notes": [], "created": "2026-08-20T09:42:33.133Z", "modified": "2026-08-20T09:42:33.256Z"}, {"entity": "publication", "iuid": "e0610b685a014764bd50b657d844f634", "links": {"self": {"href": "https://publications-affiliated.scilifelab.se/publication/e0610b685a014764bd50b657d844f634.json"}, "display": {"href": "https://publications-affiliated.scilifelab.se/publication/e0610b685a014764bd50b657d844f634"}}, "title": "Genomic regions under selection in the feralization of the dingoes.", "authors": [{"family": "Zhang", "given": "Shao-Jie", "initials": "SJ", "orcid": "0000-0003-1079-9680", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/07cf9cad475f4b3b8c8358a0ec6357b6.json"}}, {"family": "Wang", "given": "Guo-Dong", "initials": "GD"}, {"family": "Ma", "given": "Pengcheng", "initials": "P"}, {"family": "Zhang", "given": "Liang-Liang", "initials": "LL"}, {"family": "Yin", "given": "Ting-Ting", "initials": "TT"}, {"family": "Liu", "given": "Yan-Hu", "initials": "YH"}, {"family": "Otecko", "given": "Newton O", "initials": "NO"}, {"family": "Wang", "given": "Meng", "initials": "M"}, {"family": "Ma", "given": "Ya-Ping", "initials": "YP"}, {"family": "Wang", "given": "Lu", "initials": "L"}, {"family": "Mao", "given": "Bingyu", "initials": "B"}, {"family": "Savolainen", "given": "Peter", "initials": "P"}, {"family": "Zhang", "given": "Ya-Ping", "initials": "YP", "orcid": "0000-0002-5401-1114", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/08315abecebf43c0adc5874c44e3cf11.json"}}], "type": "journal article", "published": "2020-02-03", "journal": {"title": "Nat Commun", "issn": "2041-1723", "volume": "11", "issue": "1", "pages": "671", "issn-l": "2041-1723"}, "abstract": "Dingoes are wild canids living in Australia, originating from domestic dogs. They have lived isolated from both the wild and the domestic ancestor, making them a unique model for studying feralization. Here, we sequence the genomes of 10 dingoes and 2 New Guinea Singing Dogs. Phylogenetic and demographic analyses show that dingoes originate from dogs in southern East Asia, which migrated via Island Southeast Asia to reach Australia around 8300 years ago, and subsequently diverged into a genetically distinct population. Selection analysis identifies 50 positively selected genes enriched in digestion and metabolism, indicating a diet change during feralization of dingoes. Thirteen of these genes have shifted allele frequencies compared to dogs but not compared to wolves. Functional assays show that an A-to-G mutation in ARHGEF7 decreases the endogenous expression, suggesting behavioral adaptations related to the transitions in environment. Our results indicate that the feralization of the dingo induced positive selection on genomic regions correlated to neurodevelopment, metabolism and reproduction, in adaptation to a wild environment.", "doi": "10.1038/s41467-020-14515-6", "pmid": "32015346", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC6997406"}, {"db": "pii", "key": "10.1038/s41467-020-14515-6"}], "notes": [], "created": "2026-08-20T08:51:01.534Z", "modified": "2026-08-20T08:51:01.661Z"}, {"entity": "publication", "iuid": "269dc5e6c7134e14b38cf5ccbd5d661a", "links": {"self": {"href": "https://publications-affiliated.scilifelab.se/publication/269dc5e6c7134e14b38cf5ccbd5d661a.json"}, "display": {"href": "https://publications-affiliated.scilifelab.se/publication/269dc5e6c7134e14b38cf5ccbd5d661a"}}, "title": "Canine transmissible venereal tumor genome reveals ancient introgression from coyotes to pre-contact dogs in North America.", "authors": [{"family": "Wang", "given": "Xuan", "initials": "X"}, {"family": "Zhou", "given": "Bo-Wen", "initials": "BW"}, {"family": "Yang", "given": "Melinda A", "initials": "MA"}, {"family": "Yin", "given": "Ting-Ting", "initials": "TT"}, {"family": "Chen", "given": "Fang-Liang", "initials": "FL"}, {"family": "Ommeh", "given": "Sheila C", "initials": "SC", "orcid": "0000-0002-0780-0399", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/afb74fa69eef419cbca7ea9705e33133.json"}}, {"family": "Esmailizadeh", "given": "Ali", "initials": "A"}, {"family": "Turner", "given": "Melissa M", "initials": "MM"}, {"family": "Poyarkov", "given": "Andrei D", "initials": "AD"}, {"family": "Savolainen", "given": "Peter", "initials": "P"}, {"family": "Wang", "given": "Guo-Dong", "initials": "GD"}, {"family": "Fu", "given": "Qiaomei", "initials": "Q"}, {"family": "Zhang", "given": "Ya-Ping", "initials": "YP", "orcid": "0000-0002-5401-1114", "researcher": {"href": "https://publications-affiliated.scilifelab.se/researcher/08315abecebf43c0adc5874c44e3cf11.json"}}], "type": "letter", "published": "2019-07-00", "journal": {"title": "Cell Res.", "issn": "1748-7838", "volume": "29", "issue": "7", "pages": "592-595", "issn-l": "1001-0602"}, "abstract": null, "doi": "10.1038/s41422-019-0183-2", "pmid": "31160719", "labels": [], "xrefs": [{"db": "pmc", "key": "PMC6796869"}, {"db": "pii", "key": "10.1038/s41422-019-0183-2"}], "notes": [], "created": "2026-08-20T08:50:04.513Z", "modified": "2026-08-20T08:50:04.612Z"}]}
